What Is CJC-1295? | Overview

CJC-1295 is a synthetic analogue of GHRH and a growth hormone secretagogue (GHS) that was developed by ConjuChem Technologies [1]. Although it has yet to enter clinical trials, this peptide is believed to have application in the realms of anti-aging and athletic performance [2]. Despite showing promise as a research chemical, athletes beware: CJC-1295 is a prohibited substance under Section S2 of the World Anti-Doping Agency’s (WADA) Prohibited List [3].

CJC-1295 peptide is made up of 29 amino acids and is able to bind to receptors in the pituitary gland and stimulate the release of growth hormones such as plasma growth hormone (GH) and insulin-like growth factor 1 (IGF-1) [4].

Animal studies have confirmed that CJC-1295 acts as a long-lasting GRF analog and is able to activate the GRF receptors on the anterior pituitary in rats [8]. We know that CJC-1295’s potential ability to enhance performance has earned it a spot on WADA’s prohibited substances list and that it is banned from all drug-tested bodybuilding competitions. This adds weight to anecdotal claims that CJC-1295 offers similar benefits to GHRH such as increased muscle mass, fat reduction, and improvements in athletic performance.

CJC-1295 Has a Longer Half-Life and Higher Bioavailability Than GHRH 1-29

When it comes to drug efficacy and efficiency, two of the most important metrics to study are half-life and bioavailability. The half-life of a drug is a pharmacokinetic parameter defined as the time it takes for the concentration of the drug in the plasma or the total amount in the body to be reduced by 50% (half) [9]. Essentially the longer the half-life is, the longer the drug stays around in an active form.

The other piece of the puzzle is bioavailability. Bioavailability is essentially how much of the drug becomes available to its target destination [10]. If something has a low bioavailability, few benefits will be realized when administered to test subjects.

CJC-1295 DAC is a bioconjugate analogue of GHRH 1-29 meaning that is has a longer half-life and higher bioavailability due to bioconjugation, as described below [5]:

“Bioconjugation is a chemical technique used to couple two molecules together, at least one of which is a biomolecule, such as a carbohydrate, nucleic acid, or protein.

Proteins are especially diverse biomolecules due to the variety of amino acids available and are thus important substrates in bioconjugation reactions. Bioconjugation reactions play a critical role in the modification of proteins.

Because of recent advances in the study of biomolecules, proteins can be modified to perform a variety of functions, including cellular tracking, imaging biomarkers, and target drug delivery.”

CJC-1295 DAC can form bioconjugates very efficiently with albumin, a water-soluble globular protein. This reduces the rate of degradation and increases the half-life of this peptide. [8]

CJC-1295 May Promote Deep Sleep

Several studies have suggested that GHRH plays a direct role in improving the quality of sleep. In one study by Fehm et al, researchers looked at intranasal administration of GHRH and documented the effect it had on sleep quality [11]:

“A study was performed according to a double-blind cross-over design. Each of 12 young and 11 old healthy men were intranasally administered with 300 micrograms GHRH (vs. placebo) 30 min before bedtime at 23:00 h.

Sleep was recorded polysomnographically until 07:00 h and blood was collected in 15 min intervals for determination of cortisol and GH. Apart from the well-known age-related changes of hormonal secretion and sleep, intranasal GHRH reduced cortisol nadir concentrations in the beginning of sleep (P < 0.05), and also reduced the sleep-induced elevation in GH concentrations during early sleep.

Moreover, results indicated that after intranasal administration GHRH increased rapid-eye-movement (REM) sleep and slow wave sleep (SWS), with this influence concentrating on the second half of sleep time. Effects of GHRH did not depend on the subject's age.

We conclude that there is a coordinate influence of intranasal GHRH on the central nervous regulation of sleep processes and of hypothalamic-hypophysiotropic secretory activity in both young and elderly men. The effects may mimic the dual neuronal and endocrine function of hypothalamic GHRH activity.”

While this study didn’t specifically look at CJC-1295, we know this compound is an analogue of GHRH. Sleep has been linked to a myriad of benefits including tissue repair, bone and muscle density increase and a stronger immune system [12]. GH has also been shown to enhance non-REM sleep after a period of sleep deprivation [13].

CJC-1295 Increases Pulsatile GH Secretion and IGF-1 Levels

Pulsatile GH secretion is needed to get the most out of the hormone’s benefits. IGF-1 levels are also important for muscle growth. A study conducted at the University of Chicago details how CJC-1295 positively affects pulsatile GH secretion and IGF-1 levels [14]:

“CJC-1295 increased through and mean GH secretion and IGF-1 production with preserved GH pulsatility. The marked enhancement of trough GH levels by continuous GHRH stimulation implicates the importance of this effect on increasing IGF-1. Long-acting GHRH preparations may have clinical utility in patients with intact pituitary GH secretory capability.”

The interesting thing is that this increase in pulsatile GH and IGF-1 is not associated with side effects typically seen with excess GH and IGF-1 in the body.

“It is noteworthy that despite the nearly 50% increase in GH secretion and IGF-1 levels, some of which reached the upper limit of normal, none of the subjects experienced any adverse effects attributable to excessive GH or IGF-1. This may have been due to the short duration of the study, but may also reflect a protective feedback mechanism to limit the extent of CJC-1295 stimulation of GH secretion. Based on the latter assumption, the use of CJC-1295 or other GHRH analogs may confer an intrinsic advantage over that of GH as a therapeutic agent in those patients with an intact pituitary GH secretory capacity.”

These findings support claims that CJC-1295 can be used to increase the concentration of both of these important growth hormones, potentially without the side effects that typically come from having higher than normal levels of GH.

CJC-1295 May Increase Muscle Mass

Though GH and IGF-1 have both been studied in muscle anabolism, IGF-1 is more accepted as an agent of muscle hypertrophy. IGF-1 receptor activation promotes muscle anabolism by activating PI3K/Akt [15].

“Growth hormone (GH) is widely used as a performance-enhancing drug. One of its best-characterized effects is increasing levels of circulating insulin-like growth factor I (IGF-I), which is primarily of hepatic origin.

It also induces the synthesis of IGF-I in most non-hepatic tissues. The effects of GH in promoting postnatal body growth are IGF-I dependent, but IGF-I-independent functions are beginning to be elucidated. Although benefits of GH administration have been reported for those who suffer from GH deficiency, there is currently very little evidence to support an anabolic role for supraphysiological levels of systemic GH or IGF-I in skeletal muscle of healthy individuals.

There may be other performance-enhancing effects of GH. In contrast, the hypertrophic effects of muscle-specific IGF-I infusion are well documented in animal models and muscle cell culture systems.”

“The observed increase in prolactin RNA was much less marked than the increase in GH mRNA and was not dose-dependent. Nevertheless, CJC-1295 normalized prolactin mRNA levels. This increase likely reflects some degree of GHRH-induced proliferation of the mammotroph cells, previously reported in mice overexpressing GHRH.”

From these findings, it seems that CJC-1295 can also help with fat loss through prolactin regulation.

CJC-1295 Side Effects and Appropriate Dosage

Since there have been no clinical trials of CJC-1295 to date, our understanding of this compound’s side effects and appropriate dosage is limited.

We do know that in the early days of the drug’s development, ConjuChem tested CJC-1295 DAC as a potential treatment for lipodystrophy. One of the patients in the study died, though the attending physician concluded that the cause was not due to CJC-1295 [18].

The most common self-reported side effects of CJC-1295 are [

The study found that:

Based on these findings, a 75 kg test subject would require 2,250-4,500 μg (micrograms or mcg) per week.
