What is Thymosin Alpha-1?

Thymosin alpha-1 (TA-1) is a protein that consists of 28 amino acids and acts as a potent regulator of immune function [1]. It is a naturally occurring peptide fragment and was first isolated from tissue in the thymus gland, which plays the role of making T-cells and ensuring they mature properly.

T-cells are a vitally important part of the adaptive immune system as they assist it in remembering past infections and improving the function of other immune system cells. After being produced by the thymus gland, T-cells are discharged into the lymphatic system and then disperse around the body to defend against viruses and diseases.

Due to these qualities, thymosin alpha-1 has long been recognized as an immune-enhancing, immune-modulating, as well as an immune-restoring agent [2].

Thymosin alpha-1 has a pleiotropic mechanism of action, and works through toll-like receptors in both myeloid and plasmacytoid cells, thus activating signaling pathways within the immune system and stimulating the production of immune-related cytokines and other molecules that help coordinate the activities of various cells [3].

One study has suggested that the peptide alters the environment within the nervous system to favor growth and development while simultaneously blocking pathways responsible for inflammation and neuron dysfunction [10].

Recent research indicates that thymosin alpha-1 blocks angiotensin converting enzyme (ACE) and thus can reduce blood pressure.

Research has indicated that thymosin alpha-1 is well tolerated, while most studies observed only local irritation at the injection site [12]. Further, the peptide has been used for more than 40 years and has been shown to be safe by patients as well as having a low level of toxicity.

The lack of significant side effects reported with Thymosin-Alpha 1 is in sharp contrast to other major immune response modulators such as IFN and IL-2, which can lead to flu-like symptoms including malaise, fever, headache, chills and pulmonary edema (with IL-2).

Some rare side effects which have been observed with TA-1 include: Erythema, itching at the injection area, dry mouth, darkening of moles, increase of freckles, weight loss, increased hunger, and some drowsiness.

In clinical trials, thymalfasin (a synthetic TA-1 analog) when combined with interferon 2b, was reported to have rare side effects such as: fever, fatigue, muscle aches, nausea, vomiting, and neutropenia when compared to interferon-alpha 2b alone or with placebo [2].
